Synthesis of Streptococcus Pneumoniae Serotype 23F Oligosaccharide Antigen


Streptococcus pneumoniae is a deadly pathogen that can lead to meningitis, pneumonia, bronchitis, and other diseases, killing hundreds of thousands each year. Further, this bacterium recently acquired a resistance to penicillin and other antibiotics, increasing the need for vaccination. One of the most common serogroups in North America is serogroup 23, and the focus of our work has been to synthesize an effective serotype 23F polysaccharide antigen against the bacterium, allowing the human body to recognize the organism when it enters the body. The oligosaccharide vaccine matches a tetrasaccharide on the bacterium’s capsule, and the body thus has oligosaccharide-specific antibodies ready to bind to the foreign bacterium. This presentation will outline the process by which we are synthesizing the 23F tetrasaccharide antigen.
