The Effects of CMG2 on Cell Adhesion to Extracellular Matrix Proteins

Abstract

Capillary Morphogenesis Gene 2 (CMG2) is a transmembrane protein that is upregulated during angiogenesis. It also acts as an anthrax toxin receptor and animals treated with anthrax protective antigen (PA) show reduced angiogenesis. Hence, CMG2 has significant potential as a target for anti-angiogenic therapy in the treatment of cancer and eye diseases.  However, the full extent of its physiological role is poorly understood. Through adhesion assays, using PA as a high affinity inhibitor of CMG2, we have shown that CMG2 promotes endothelial cell adhesion to various matrix proteins.  These results suggest that CMG2 mediates cell adhesion through extracellular matrix and helps establish the physiological role of CMG2. In addition, we have shown that a small fragment of collagen IV (S16) inhibits endothelial cell migration and is a promising approach for new anti-angiogenic therapies.
