Ceragenins in the Respiratory System: An Ev vivo Study

Abstract

Patients with cystic fibrosis (CF) are prone to experience chronic lung infections due to the accumulation of mucus in their lungs. Ceragenins are mimics of endogenous antimicrobial peptides that are promising candidates as a treatment option for CF patients due to their broad-spectrum antimicrobial activity and ability to eradicate biofilms. However, past studies have shown many antibiotics to have harmful effects on cilia, which are necessary for the removal of particulate matter from the lungs and trachea. The harmful impact of ceragenins on cilia would limit their use as antimicrobials. In attempt to mitigate cilia damage we have combined ceragenin CSA-131 with the polaxamer Pluronic F127. The effects of this combination were studied in an ex vivo porcine model. In a bead clearance assay on ciliated epithelial tissues the combination was shown to prevent cilia damage. Additionally, efficacy tests showed the ceragenin retained significant antimicrobial activity when combined with the pluronic.
