Formation of Thrombin-Antithromin Complex for Separation of Preterm Birth Biomarkers


Preterm birth (PTB) is the leading cause of neurological disabilities and infant mortality. Over one million infants died in 2013 from PTB related complications. Esplin, et al.1 found that PTB risk can be determined with 87% selectivity and 81% specificity through nine maternal blood serum biomarkers. One of these biomarkers is the thrombin-antithrombin III complex (TAT). TAT cannot be purchased so it must be synthesized in the lab to be analyzed. We have formed TAT in buffer and analyzed the results by dot blot test, capillary electrophoresis, and microchip electrophoresis. Once synthesized, we were able to separate TAT from six other PTB biomarkers using microchip electrophoresis. We are presently working to integrate this separation with upstream microfluidic sample preparation steps for the diagnosis of PTB risk.
