Degradation of Processing Bodies is Mediated by LIMD1-Dependent Autophagy


LIM domain containing protein 1 (LIMD1) is a scaffolding protein that was identified as a tumor suppressor in 2007.  We hypothesize that LIMD1 recruits Atg9A and 14-3-3ζ to P-bodies to promote the selective degradation of P-bodies. This degradation of P-bodies may lead to the degradation of P-body-associated oncogene mRNA transcripts.  We identified the autophagy-regulating proteins Atg9A and 14-3-3ζ as binding partners of LIMD1 and identified a phosphorylation on LIMD1 (pS316) that is essential for 14-3-3ζ binding. We are currently using a phospho-null LIMD1 mutant (S316A) to determine the role of 14-3-3ζ in mediating the LIMD1-ATG9A interaction and P-body degradation. Together, these aims will help to uncover another tumor suppressing role of LIMD1 that may be used therapeutically to target cancer progression.
