Title

Decellurization of Porcine Hearts for Pharmaceutical Research

Abstract

Heart disease is the leading cause of death in the United States. During myocardial infarction cardiac tissue suffers a lack of nutrients and oxygen that leads to the formation of unregenerable scar tissue which causes a loss of myocardial functionality. With the advent of human induced pluripotent stem cells (hiPSC), research has greatly increased in engineered cardiac tissues (ECTs) as a translatable treatment to such loss of cardiac function and as a model for pharmaceutical research. We create ECT's using hiSPC differentiated into cardiomiocytes on the extracellular matrix derived from the left ventricle of a decellurized porcine heart. The decellurization process allows for the porcine cells to be denatured and subsequently washed from the heart tissue. It begins with a phosphate-buffered saline solution (PBS) wash to remove blood and other fluids present in the heart in an isotonic environment. Then there is a deionized water rinse, followed by perfusing the heart with sodium dodecyl sulfate. This denatures cell membranes, lipids, sugars, and DNA, leaving behind the insoluble collagen fibers of the extracelluar matrix. Triton X 100 completes the process.
