Application of the Intramolecular Styryl Diels-Alder reaction (ISDA) to Construct the Anticancer Agent Deoxypodophyllotoxin DPT and Antiviral F4

Abstract

The intramolecular styryl Diels-Alder reaction (ISDA) is a [4+2] cycloaddition with immense value in organic synthesis. Three rings and one stereocenter can be constructed in a single step making the core structure of some valuable natural products. Limited examples of this methodology have been published due to major drawbacks which include loss of aromaticity and problematic thermal [1-3] hydrogen shift. Besides those drawbacks, we have successfully applied the ISDA reaction in the construction of racemic Deoxypodophyllotoxin (DPT). This talk will present our analysis of the ISDA reaction as well as our synthetic routes for DPT and F4.
