Analysis of Thrombin-Antithrombin Complex Formation using Microchip Electrophoresis and Mass Spectrometry to Facilitate Preterm Birth Risk Diagnosis

Abstract

Preterm birth (PTB) is a major cause of infant mortality and disabilities. We are working to assess PTB risk through analyzing nine maternal blood biomarkers. We previously developed a separation method for six of these biomarkers via microchip electrophoresis (μCE).1 However, the thrombin-antithrombin III complex (TAT) was omitted from this method, because it is not available commercially and thus must be formed in the lab to be analyzed. We developed a method for TAT formation and validated it using dot blots and MS-MS. We also evaluated TAT formation by μCE comparisons of TAT and its components. Moreover, fluorescent labeling conditions for TAT were determined to maximize its μCE signal. Finally, μCE separation of TAT with other PTB biomarkers was demonstrated.
